During this project a novel, inducible baculovirus-insect cell-expression system able to produce cell toxic proteins and high virus titers shall be developed and extensively tested.
For this purpose a two-promoter-system shall be used , in which the transcription of the GOI is controlled by a strong constitutive promoter , while at the same time post-transcriptional gene-silencing is regulated by an inducible second promoter .
This allows selective turning on and off of toxin expression and therefore facilitation of regulation of the protein production process.

Hierzu soll ein Zwei-Promotor-System verwendet werden, in dem die Transkription des GOI durch einen starken konstitutiven Promotor kontrolliert wird und gleichzeitig durch einen induzierbaren Promotor das post-transkriptionelle Gen-Silencing reguliert werden kann.
So lässt sich die Expression toxischer Proteine gezielt an- und ausschalten, wodurch der Produktionsprozess deutlich leichter gesteuert werden kann.
